First Report of Cucumber Mosaic Virus Natural Infection in Euphorbia Epithymoides in Ukraine

Ornamental plants grown in urban and peri-urban environments may act as persistent CMV reservoirs, facilitating virus maintenance and spillover into nearby agricultural crops. Although Cucumber mosaic virus (CMV) is known to infect a wide range of ornamental plants, including species of Euphorbia, its occurrence in Euphorbia epithymoides has not previously been documented in Ukraine. In June 2021, cushion spurge plants (Euphorbia epithymoides L.) exhibiting virus-like ringspot symptoms were spotted in Kyiv, Ukraine. This study aimed to identify potential virus agents damaging Euphorbia plants using serological and molecular methods. Methods. Symptomatic and healthy plants were tested for multiple viruses using ELISA, followed by RT-PCR and sequencing. Phylogenetic analysis was conducted to determine the genetic relationship of the detected virus. Results. CMV was serologically identified in 7 of 9 symptomatic plants, and confirmed via RT-PCR and sequencing. To rule out mixed infections, the samples were also tested for 73 potyviruses, as well as for tobacco mosaic virus, tomato mosaic virus, tobacco rattle virus, alfalfa mosaic virus, arabis mosaic virus, tomato spotted wilt virus, impatiens necrotic spot virus, and tobacco ringspot virus. Apart from CMV, no other tested viruses were detected. Phylogenetic analysis revealed that the CMV isolate belongs to subgroup II, with 98.72% nucleotide identity to isolates from Slovakia, Greece, and Japan. Conclusions. This study is the first report on natural infection of E. epithymoides by CMV, expanding knowledge of the virus’s host range and genetic diversity.
